Streptomycin works on 30S subunit; puromycin is analog of 3’ aa-tRNA end.
Puromycin adds to A site without EF-Tu or EF1A and accepts the peptidyl chain from the P site peptidyl-tRNA: dead end product, because the peptidyl chain is linked via and amide bond to the 3’ NH instead of the usual 3’ OH found in tRNAs.
